DESCRIPTION
JC-10 is a novel fluorescent indicator that can be utilized to monitor mitochondrial membrane potential. It has several key advantages over earlier generations of indicators like JC-1:

JC-10 is a cationic, lipophilic dye that can be used to monitor mitochondrial membrane potential. When mitochondrial membrane potential is low, JC-10 emits a green fluorescence. Conversely, when mitochondrial membrane potential is high, JC-10 will emit a green-orange fluorescence.
Assay Principle
JC-10 is a cationic, lipophilic dye that can be used to monitor mitochondrial membrane potential. When mitochondrial membrane potential is low, JC-10 emits a green fluorescence. Conversely, when mitochondrial membrane potential is high, JC-10 will emit a green-orange fluorescence.

The shift in emission is a result of JC-10’s unique ability to reversibly form aggregates as a function of mitochondrial membrane potential. At low membrane potentials, JC-10accumulation in mitochondria is low and most of the JC-10will be in the monomeric form, which will emit at ~520 nm when excited by ~490 nm light source. At high membrane potentials, JC-10 accumulation in mitochondria is high and most of the JC-10 will be in the aggregate form, which will emit of ~570 nm when excited by ~490 nm light source. Because JC-10 aggregate formation is a function of membrane polarization, the fluorescence intensity of JC-10 aggregates reflects the degree to which a mitochondrial membrane is depolarized or hyperpolarized.
While there are older probes that can monitor mitochondrial membrane potential, such as JC-1, Celltechgen™ JC-10 Dye outperforms them in several key aspects:
- Unlike JC-1, which tends to precipitate even at low concentrations (ie. 1µM), our Celltechgen™ JC-10 Dye is much more soluble in aqueous solutions. This allows for easier preparation and handling.

JC-10 can be used to study mitochondrial membrane potential in a wide array of cell types such as neurons and myocytes, and can be used to study critical cell processes such as ATP synthesis, reactive oxygen species (ROS) and apoptosis.
