Description: Approximately 50.5 kDa. Recombinant Protein A/G consists of 7 IgG-binding domains EDABC-C2C3, which corresponds to Protein A and G domains that are included in the recombinant sequence. Protein A portion is from Staphylococcus aureus segments E, D, A, B and C. Protein G portion is from Streptococcus segments C2 and C3. The binding capacity of recombinant Protein A/G is broader than either Protein A or Protein G alone.
Purity: >95% by SDS-PAGE analyses.
Specificity: The extended Fc-binding properties of Protein A/G make it a popular tool in the investigation and purification of immunoglobulins. Protein A/G binds to all human IgG subclasses, IgA, IgE, IgM and to a lesser extent IgD; however, it does not bind mouse IgA, IgM or murine serum albumin.
Applications: Protein A/G is an excellent tool for purification and detection of mouse monclonal antibodies from IgG subclasses without interference from these other serum proteins. Individual subclasses of mouse monoclonals are most likely to have stronger affinity to this chimeric protein than to either Protein A or Protein G. Binding is less pH-dependent than either Protein A or Protein G alone, occurring well at pH 5-8.
Formulation: Lyophilized from a 0.2μm filtered solution of 20mM PBS, pH 7.0, containing 2% mannitol.
Endotoxin: Less than 10EU/mg of rhirudin as determined by LAL method.
Reconstitution: We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL. Stock solutions should be apportioned into working aliquots and stored at <-20°C.
Storage: This lyophilized preparation is stable at 2-8°C, but should be kept at -20°C for long term storage, preferably desiccated. Upon reconstitution, the preparation is stable for up to one week at 2-8°C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20°C to -70°C.Avoid repeated freeze/thaw cycles.
